Cryopreserved human hepatocytes culture optimization on polymethylpentene oxygen permeable membranes for drug screening purposes

چکیده

In vitro culture of primary hepatocytes for drug screening purposes remains a challenge as cells rapidly lose their function in conventional conditions. Thin oxygen permeable membranes have shown, through direct oxygenation, beneficial effects long-term and cellular with freshly isolated hepatocytes. However, cryopreserved hepatocytes, standard the industry, has shown limits due to high damage, leading low function. addition, sorption compounds on PDMS rendered evaluation different molecules, aimed at improvement those difficult. Here, was performed PMP membranes, known exhibit exceptionally characteristics. A mixture anti-apoptotic anti-inflammatory improve cell viability during adhesion tested terms damage metabolism after 24 hr 72 hr. Components were effect Reactive Oxygen Species production 6 well mitochondrial activity LDH release Effects improving recovery albumin metabolism, could be efficiently measured result use PMP. The presented results demonstrate compatibility oxygen-permeable membrane-based efficient screening.

A novel in vitro experimental system for the evaluation of drug metabolism: Cofactor-supplemented permeabilized cryopreserved human hepatocytes (MetMax™ Cryopreserved Human Hepatocytes).

We report here a novel experimental system - MetMax™ cryopreserved human hepatocytes (MMHH), for in vitro drug metabolism studies. MMHH consist of cofactor-supplemented permeabilized cryopreserved human hepatocytes. The use procedures for MMHH are significantly simplified from that for conventional cryopreserved human hepatocytes (CCHH): 1. Storage at -80o C instead of in liquid nitrogen; 2. Us...

Oxygen permeable ceramic membranes for hydrocarbon conversion reactors

Because of their various potential applications, non-porous, selective oxygen permeable ceramic membranes with mixed ionic and electronic conductivity (MIECs) have received significant interest over the past decade [1-8]. In particular, the incorporation of these materials into catalytic reactors for the oxidation of hydrocarbons is being investigated as a way to eliminate costly air separation...

Evaluation of Adverse Drug Properties with Cryopreserved Human Hepatocytes and the Integrated Discrete Multiple Organ Co-culture (IdMOCTM) System

Human hepatocytes, with complete hepatic metabolizing enzymes, transporters and cofactors, represent the gold standard for in vitro evaluation of drug metabolism, drug-drug interactions, and hepatotoxicity. Successful cryopreservation of human hepatocytes enables this experimental system to be used routinely. The use of human hepatocytes to evaluate two major adverse drug properties: drug-drug ...

Comparison of cryopreserved HepaRG cells with cryopreserved human hepatocytes for prediction of clearance for 26 drugs.

Prediction of clearance in drug discovery currently relies on human primary hepatocytes, which can vary widely in drug-metabolizing enzyme activity. Potential alternative in vitro models include the HepaRG cell (from immortalized hepatoma cells), which in culture can express drug-metabolizing enzymes to an extent comparable to that of primary hepatocytes. Utility of the HepaRG cell will depend ...
